Fifteen cards that are almost an exact fit into the compartments.
Five of these cards shown at "A" have a picture of Jesus on the face and one of the letters of the word J E S U S on the back.
Five of the cards at "B" are matt black to match the card stand compartments.
The remaining five cards "C" have pictures on one side.  The first is a holiday picture the second is a picture of a school etc.

Set-up
Four of the JESUS cards are placed into the recesses on the stand and then covered with the black cards.  The fifth black card is placed into the vacant space, (we will call this space 1).

The other five cards including the last JESUS card with the S on it's back are on your table or in your pocket.

Presentation
Place the five picture cards into the spaces on the stand.  You can show the backs of these cards but don't make a point of it.

Show the Jesus card and ask your audience where they would most likely expect to find Jesus.  The reply will almost certainly be "In Church".

Place the Jesus card behind the church card.

Ask the audience "what happens when everyone goes home? is Jesus left there all alone in the church?"

Remove the church card to reveal the Jesus card still there.  Take this out and replace the church card.

Tilt the church card forward as well as the black card behind it and put the Jesus card back into the stand behind both of the cards (The Jesus card is now behind the black card).

Continue with your patter. "No when you go home, Jesus goes with you".

Lift the church card out and the audience will just see the black card covering the Jesus card.  Replace the church card and lift out the house or home card taking the black card with it (this is a double lift). This will reveal the Jesus card that is in that position.

By repeating these moves the Jesus card can be made to vanish and re-appear in any of the spaces.

Finish by saying that "Jesus is with us all wherever we are".

Remove all the picture cards and black cards (double lifts) to show a Jesus card at each position.

Climax the effect by asking "Who do you think my best friend is?"

As you say this turn all the Jesus cards over in their spaces so that they spell out the name
J E S U S.
